Effect of Purified Curcuminoids on Enterococcus faecalis and Its Biofilm

Abstract
Objective: Curcuminoids is a group of natural compounds extracted and purified Tumeric rhizome (Curcuma longa Linnaeus). The objective of this study was to investigate the effects of its antimicrobial and antibiofilm properties on Enterococcus faecalis, a harsh pathogen for oral disease.

Methods: Purified curcuminoids was tested for antimicrobial activity against E. faecalis by microdilution assay (minimal in-hibitory concentration [MIC] and minimal bactericidal concentration [MBC]). Its antibiofilm characteristic was also measured on 96-well microtitre plate using crystal violet staining and colony forming unit counting techniques. To study the influence of curcuminoids in molecular level, gelatinase was chosen as a main component closely related to biofilm formation. Enzyme hydrolysis test was conducted and the expression of a relevant gene: gelE (gelatinase) was performed by semi-quan-titative reverse transcription polymerase chain reaction.

Results: Curcuminoids showed antimicrobial activity against E. faecalis, with MIC of 25 ?g/ml and MBC of 50 ?g/ml for planktonic cells, whist with MIC of 50 ?g/ml and MBC of 500 ?g/ml for biofilm forming cells. The inhibitory effect on bio-film formation was shown by the reduction of biofilm mass with the minimal concentration of 37.5 ?g/ml. Gelatinase activity was also obviously inhibited. The gelE (gelatinase) expression was found down-regulated after 24 hours exposure of E. faeca-lis to purified curcuminoids.

Conclusion: Curcuminoids has abilities to inhibit biofilm formation and growth of E. faecalis. These characteristics involve the mechanism of biofilm formation relating to gelatinase enzyme and gelE (gelatinase) of E. faecalis.
